Effects of acute and prolonged administration of benzydamine on the stimulatory effect of ethanol in mice (2000)

- Abstract: Objetivo:The purpose of the present study is to evaluate the acute and prolonged effect of benzydamine (BEZ) on the locomotor activity and to verify whetther stimulatory effect ethanol (EtOH) is modified in BEZ.Métodos e Resultados: In acute experiments, separate groups of male Swiss mice were treated with BEZ (15, 30, or 60 mg/kg. i.p., single dose) or saline (SAL) 30 min before the injections of EtOH (2.0 mg/kg, i.p.) or SAL. In prolonged experiments, separate groups of mice (N=8) were injected once a day with BEZ (15 mg/kg, i.p.) for 21 consecutive days. All animals recieved SAL for the next 2 days. On the third day after discontinuation of the drug treatment, mice recieved a challenge dose of EtOH (2.0 mg/kg, i.p.). Thirty minutes after the EtOH injection the evaluation of locomotor activity was recorded as described above. BEZ (60 mg/kg) significantly decreased EtOH-induced locomotion (SAL + EtOH: 211 ± 11; BEZ60 + EtOH 139 ± 9). The prolonged administration of BEZ (15 mg/kg/day) produced a significant increase in EtOH-induced locomotion (SAL + EtOH: 171 ± 11; BEZ15 + EtOH 229 ± 25).Conclusões: Our present data showed that the repeated exposure to BEZ intensified the EtOH-induced on stimulatory effect. This observation can be interpreted as behavioral sensitization or cross-sensitization
