Ovarian hormones modulates endothelin-1 (ET-1) vascular reactivity in DOCA-sal hypertensive rats (2001)

- Abstract: Objective: Since male, but not female, DOCA-salt rats exhibit marked vasoconstriction upon ET-1/ETB agonist stimulation, we hypothesized that ovarian hormones attenuate ETB responses in female DOCA rats. Wistar rats underwent ovariectomy (OOF) and received sc pellets containing estradiol (E) or E+progesterone (EP). Design/Methods: After 2 weeks, rats were randomized in 6 groups: control, control-OOF, DOCA, DOCA-OOF, DOCA-OOF+E, DOCA-OOF+EP. Responses to IRL-1620 (IRL), an ETB agonist, were evaluated in vivo/in situ in the mesenteric microcirculation. ET-1 and ETB mRNA expression was evaluated in mesenteric arteries by RT-PCR and expressed relatively to GAPDH. Results: DOCA-OOF rats developed a more severe form of hypertension than DOCA rats (186±4 vs 164±3 mmHg). DOCA-OOF treated with E or EP exhibited lower blood pressure (BP) levels (170±3 vs 173±4 mmHg). OOF did not change BP or IRL responses in control-OOF rats. In the mesentery, IRL 10-9M induced vasodilatation in control (% change initial diameter, +7.4±1%), a mild vasoconstriction in DOCA (-4.5±2%) and marked vasoconstriction in DOCA-OOF (-9.8±1%) rats. E restored IRL-induced vasodilatation (+4.9±1%). ET-1 mRNA was increased in arteries from DOCA-OOF (2.2±0.1), but not DOCA (1.4±0.2) or DOCA-OOF+E (1.5±0.3) vs control (1.0±0.2) and control-OOF (1.4±0.2). ETB mRNA levels were greatly enhanced in DOCA-OOF (1.6±0.4) vs DOCA (0.9±0.1), DOCA-OOF+E (0.6±0.3) and control (0.6±0.1) arteries. Conclusions: These resultssupport our suggestion that the ovarian hormones, specially estradiol, modulate vascular responses to ET-1/ETB agonists in DOCA-salt hypertension
