The role of cyclic AMP-dependent protein kinase and nitric oxide-cyclic GMP and sodium, potassium-atpase in behavioural sensitization by fencamfamine (2002)

- Abstract: Fencamfamine (FCF) is a Central Nervous System (CNS) stimulant that facilitates dopaminergic transmission through blockade of dopamine (DA) uptake and in part by increasing DA release from nerve terminals. The repeated administration of FCF and other amphetamine(AMPH)-like stimulants result in an altered behavioral response to the subsequent administration of the drug. Although the exact neural mechanisms involved in the behavioral sensitization have not been identified, several authors point to a function of GLU in the development of sensitization to AMPH and cocaine. The GLU-nitric oxide (NO) modulatory action on cyclic GMP and Na,K-ATPase activity were evaluated in the nucleus accumbens (NAc) of animals acutely or repeatedly treated with fencamfamine (FCF - 3.5 mg/kg). Behavioral sensitization occurs in long-term treated animals which was linked to an increase in NO-synthase activity (16.3 ± 0.6 pmol/mg protein.min) when compared to acute (12.3 ± 0.3) and control groups (11.3 ± 0.8). On sensitized animals there was also an increase in cyclic GMP levels (6.7 ± 0.8 pmol/mg protein) comparing to the acute (4.7 ± 0.7) and control (4.5 ± 0.8) groups. Na,K-ATPase showed a similar activity in control (138.3 ± 19.4 nmol of Pi/mg protein.min) and sensitized animals (132.5 ± 18.3), but was reduced in acute treated rats (51.2 ± 13.4). These results led us to suggest that sensitization induced by FCF is probably linked to a modification in Na,K-ATPase activity throughNO-cyclic GMP, leading to changes in neuronal excitability related to pharmacological stimulation on the NAc
