Alterations in muscle mass and mhc pro-file induced by immobilization: effect of creatine supplementation (2003)

- Abstract: Objetivo: Hypokinesia induced by immobilization drives important changes in the skeletal muscle, such as, severe mass loss and fiber type shift. Since creatine is involved in skeletal muscle tropism, our aim was to investigate the effect a high dose of creatine feeding (5 g.kg-1 body weight.day-1) upon muscle plasticity during hind limb immobilization. Métodos e Resultados: To access that hypothesis Wistar rats were submitted to hind limb immobilization in association to 3 dietary manipulations: control (C n=6), supplemented with creatine during immobilization (Cre n=6) and pre-supplemented with creatine for 7 days before and during immobilization (PCre n=6). The left leg was immobilized (I) in shortening position and the contra-lateral leg remained lose (CL). Muscle weight (wet and dry) was determined in soleus (Sol), gastrocnemius (Gas) and extensor digitorium longus (EDL). The analysis of the lean mass was performed by DEXA and MHC distribution by SDS-PAGE. Statistical analysis was performed by ANOVA (Tukey test, p<0.05).) Results: Regardless dietary manipulation (C, Cre and PCre), the immobilization procedure decreased relative wet weight of Sol (by 35, 30 and 20%, respectively) and Gas (by 28, 25 and 20%, respectively). The leg lean mass also displayed marked decrease by the immobilization in all groups. Creatine feeding performed before and during immobilization reduced muscle mass loss comparing to C in both muscles (Sol - C: 0.33±0.02,PCre: 0.42±0.02*; Gas - C: 3.91±0.15, PCre: 4.63±0.13*) (*p<0.05). Our findings also indicate that creatine supplementation maximizes the expected slow-to-fast MHC shift observed during immobilization (Immobilized Sol- MHCIIa - C: 15.9±1.1, Cre: 21.8±1.4*, PCre: 20±1.2*) (*p<0.05). Conclusões: Previous creatine supplementation attenuates the deleterious effects of immobilization in muscle mass, although it further enhances the fast twitch fiber phenotype induced by hypokinesia
