Ultrastructure of the alveolar bone-implant interface in rat mandibles (2003)

- Abstract: Objectives: In order to evaluate the interaction between bone and implants, many authors have used animal models, mainly in the tibia from rats or rabbits. Although bone possesses a similar structure at the several regions of the body, the alveolar bone exhibits some particularities. Thus, the present investigation aims an ultrastructural study in order to analyze morphological aspects of alveolar bone after installation of titanium mini-implants in rat mandibles. Methods and Results: One screw-type mini-implant (1.4mm of length x 1.2mm of width) was placed between the mesial and distal buccal roots of the lower first molar from fifteen Wistar rats. The animals were killed after 45, 60 and 90 days and the mandibular portion containing the first molar and the implant was removed and fixed in 2% glutaraldehyde + 2.5% formaldehyde buffered with 0.1M sodium cacodylate pH 7.4, under microwave irradiation.Then, specimens were decalcified in 4.13% EDTA, washed, dehydrated and embedded in Spurr resin. The blocks were cut, the mini-implants removed and the fragments embedded in resin again. Ultrathin sections were analyzed in a JEOL 1010 electron microscope. The ultrastructural analysis at the interface between the alveolar bone and the implant revealed evident bone formation in all the specimens. After 45 days, the implant surface was covered by connective cells, which were surrounded by a collagen-rich organic matrix. At more advancedtimes, some regions of the interface exhibited flattened cells, which appeared to be entrapped against the implant. In other regions, the bone tissue reached the implant surface, exhibiting some osteocytes close to the interface. Conclusion: The bone formation observed in contact with the surface of all the implants revealed that osseointegration took places. This model, therefore, besides facilitating the tissue processing for ultrastructural studies, allows the investigation of the relationship between titanium implants and other periodontal tissues
