The role of nitric oxide and oxidative status in the reduction of NaK-ATPase activity in brain induced by chronic inpredictable stress (USt) (2003)

- Abstract: Long-lasting stress can have numerous pathologic effects ranging from minimal behavioral alterations to neurodegeneration. It has been shown that stress increases the neuronal release of glutamate, alters the intracellular level of Ca2+ and depletes the energy stores in the brain. The enzyme NaK-ATPase has a fundamental role in the maintenance of ionic homeostasis and restoring the resting potential in neurons. We investigated whether USt modulates NaK-ATPase activity in the prefrontal cortex (FCx) and the possible role of oxidative/nitrosative status in this regulation. Adult male Wistar rats (n=6) were exposed to USt for 14 days and in the 15th day, both control (C) and USt animals were sacrificed and FCx was removed. USt decreased NaK-ATPase activity in FCx (22%, p<0,05) but not the expression of the NaK-ATPase isoforms. Interestingly, an increase of protein nitration was observed in FCx of USt rats when analyzed by Western blot.Furthermore, when compared to C, USt increases inducible nitric oxide synthase (NOS) activity (250%, p<0.05) as well as malondialdehyde (an indicator of lipid peroxidation) levels in FCx (56,8%, p<0.05) without changes neuronal NOS activity. These findings suggest that an increase in oxidative status accounts for the reduction in NaK-ATPase activity in brain after stress
