Lumphocyte infiltration in a murine asthma model is modulated by nitric oxide (2003)

- Abstract: Objective: The effect of a nitric oxide synthase inhibitor was studied in a mouse (C57Bl/6) model of allergic lung inflammation. Methods and results: Nitric oxide synthase inhibitor, Nw-nitro-L-arginine methyl ester (L-NAME) and the inactive enantiomer, Nw-nitro-D-arginine methyl ester (D-NAME) was given i.p. to ovalbumin sensitized mice, 30 min before antigen challenge. After 24 h, bronchoalveolar lavage was perfomed for cell analysis and the lungs were removed for evaluation of airway reactivity to metacholine and mucus production. Treatment with L-NAME caused a significant decrease in the number of lymphocytes CD4+ (58.84%), CD8+ (59.71%), B220 (66.67%), Tgd+ (52.46%) and NK1.1+ (71.09%) in the bronchoalveolar lavage fluid. Mucus secretion was significantly reduced (36%) by this treatment and airway hyperreactivity was abolished. Conclusion: These results show that nitric oxide produced in allergic lung inflammation in mice contributes to the infiltration of lymphocytes (CD4+, CD8+, B220, Tgd+ and NK1.1+), airway hiperreactivity and mucus production
