The expression of substance P MRNA in the rat spinal cord submitted to a contusion lesion (2005)

- Abstract: Objective: The neuropeptide substance P is a neurotransmitter in the central nervous system. The molecule also participates in the neuroplasticity of the spinal cord, especially after injury. Methods and Resulted: Adult rats were submitted to a moderate spinal cord injury by dropping a 10g rod (25mm distance) using the NYU Impactor (Lesion Group). A group of rats was submitted to a sham operation (Sham Group). The animals were sacrificed 3, 7 or 45 days after the surgery and their spinal cords removed. The substance P mRNA signal was achieved in the spinal cord by means of radioactive in situ hybridization and quantified using microdensitometric analysis (optical density). The expression of substance P mRNA signal was increased in anterior horn at thoracic level close to the lesion epicenter [0.047±0.013 (Sham) and 0.119±0.022 (Lesion), p<0.05] 45 days after spinal cord injury. At high thoracic level, the signal in central intermediate gray matter was decreased 7 days post lesion [0.254±0.014 (Sham) and 0.154±0.025 (Lesion), p<0.05], but was increased 45 days after spinal cord injury [0.049±0.009 (Sham) and 0.135±0.03 (Lesion), p<0.05]. A 24% increase was found in laminae IV of the posterior horn at cervical level after 45 days of contusion and a 34% increase was observed in laminae I and II of the gray matter at thoracic level by 3 days. Conclusions: Substance P is synthesized in the spinal cord and may be involved in the local plasticity after lesion
