Role of apob-derived peptides on oxidized low density lipoprotein (OX-LDL)- induced endothelium dysfunction (2005)

- Abstract: Objetivo: Oxidative modification of LDL plays a pivotal role in atherosclerosis. Ox-LDL is a heterogeneous population of particles and molecules and little is known whether some of the described atherosclerosis-related properties of oxLDL in smooth muscle cells, macrophages and endothelial cells could be attributed to a defined constituent. In this study we investigated the effects of Apolipoprotein B (apoB)-derived peptides on the vascular reactivity of Wistar rats. Métodos e Resultados: Concentration-response curves to acetylcholine (ACh), sodium nitroprusside (SNP) and noradrenaline (NA) were performed in aortic (Ao) and mesenteric (Mes) arterial rings. Vessels incubated with oxLDL were less sensitive (Ao, n=5: logCE50= -7.1±0.05M vs. control -7.3±0.05M, p<0.05; Mes, n=6: logCE50= -6.9±0.08M vs. control -7.3±0.12M, p<0.05) and displayed decreased maximal response (Ao= 62.1±8% vs. control 87.1±2%, p<0.05; Mes= 66.9±7% vs. control 83.2±3%, p<0.05;) to ACh. ApoB-derived peptides A, B and C also decreased ACh responses (p<0.05) both in Ao and Mes arteries (Ao maximum relaxation, peptide A: 68.5±7%, B: 75.1±1%, C: 75.4±4% vs. control 89.9±2%; Mes, peptide A: 72.6±4%, B: 53.0±9%, C: 48.4±9%, vs. control 88.8±9%). Neither ox-LDL or the ApoB-derived peptides changed responses to NA or SNP. Conclusões: We demonstrate that fragmentation of the apoB protein can generate peptides that may contribute to endothelial and vascular dysfunction in atherosclerosis
