Recombinant expression of antimicrobial peptides from the cattle tick Boophilus microplus (2005)

- Abstract: Objetivo: In this work, we propose the obtention of recombinant peptides for ixodidin and microplusin, both isolated from hemocytes and cell-free hemolymph of B. microplus,respectivelly and analyse their mode of action. Métodos e Resultados: Previous studies showed that ixodidin is a 7,103 Da AMP, with tem cysteine residues and bacterial properties against Micrococcus luteus and proteolytic inhibitory activity against elastase and chymotripsin (Fogaca et al., abstract presented in this meeting, 2003). After unsuccessful tentatives to clone the ixodidin cDNA by RT-PCR, we decide construct a synthetic gene from the deduced sequence of aminoacids, where a fragment, corresponding to the N-terminal portion of the ixodidin was synthesized by elongation of the two annealed oligonucleotides and other two oligonucleotides were used to synthesize a fragment, corresponding to the C-terminal portion of the peptide. After ligation of the two fragments, the complete gene was cloned in the yeast vector pTG 4812 and expressed in Sacharomycies cerevisiae W303. The other peptide, microplusin, has 10,204 Da, six cysteine residuesand bacteriostatic effect on M. luteus (Fogaca et al, Develop. Comp. Immun. 28: 191-200, 2004). The previously cloned cDNA was subcloned and expressed as described for ixodidin. Conclusão: Presently, the recombinant AMPs are being purified by cromatography for future studies about their biological activity and mode of action
