Nitric oxide synthase, angiotensin AT1- and AT2- receptors and bradykinin B2-receptor in male and female spontaneously hypertensive rats treated with losartan (2005)

- Abstract: Objective: We demonstrated that losartan corrects the impaired endothelium-dependent vasodilation in mesenteric arterioles more easily in female than in male spontaneously hypertensive rats (SHR), (J Hypertension, 22:S176, 2004). In attempt to explain this difference, we investigated the effect of losartan on the nitric oxide synthase (NOS), angiotensin AT2- and AT1-receptors and bradykinin B2-receptor. Methods and Results: Male and female SHR (14-16 weeks) were divided into 03 groups (3-7 animals each) treated or not with losartan for 24 hours or 15 days (15mg/kg/day). Mesenteric vascular bed was dissected and NOS activity was evaluated using radiolabelled L- [3H]arginine conversion to L-[3H]citrulline (pmol/mg.min). Expression of eNOS, AT1- and AT2-receptors, and B2-receptor were evaluated by RT-PCR. Chronic or acute losartan treatment did not alter NOS activity in male (5.6±0.8 untreated vs 7.0±0.3 24h vs 3.9±0.4 15d) and female (5.9±0.8 untreated vs 6.2±0.3 24h vs 4.3±0.8 15d) SHR. eNOS and AT1- receptor expressions were not modified by losartan treatment in male and female. B2-receptor expression was increased after acute or chronic treatment with losartan either in male or female SHR. AT2-receptor expression was increased only in female after acute or chronic treatment with losartan. Conclusions: Correction of the arteriolar dilatation in SHR may involve increase in the expression of bradykinin B2-receptor. Angiotensin AT2 receptor expression could accountfor the differences in the responses of male and female SHR to losartan treatment
