Endothelin-1 induces neutrophil recruitment in adaptive inflammation via TNF'alfa' and CXCL1/CXCR2 in mice (2012)

- Abstract: L’endothéline médie le recrutement des neutrophiles durant l’inflammation innée. Dans la présente étude, on a examiné si l’endothéline-1 (ET-1) joue un rôle dans le recrutement des neutrophiles dans l’inflammation adaptative chez les souris, ainsi que les mécanismes associés. On a utilisé des traitements pharmacologiques pour déterminer le rôle de l’endothéline dans le recrutement des neutrophiles dans la cavité péritonéale de souris provoquées avec un antigène (ovalbumine) ou l’ET-1. On a déterminé les taux d’ET-1, TNF-‘alfa’ et de CXCL1 par ELISA, et effectué l’analyse de la migration des neutrophiles de même que l’analyse en cytométrie en flux des neutrophiles 4 h après le stimulus intrapéritonéal (i.p.). L’ET-1 a induit un recrutement dose dépendant des neutrophiles dans la cavité péritonéale. Les traitements avec l’antagoniste non sélectif des récepteurs ETA/ETB, bosentan, et les antagonistes sélectifs des récepteurs ETA et ETB, BQ-123 ou BQ-788, respectivement, ont inhibé la migration des neutrophiles induite par l’ET-1 et par l’ovalbumine dans la cavité péritonéale. De même, la provocation avec l’antigène a augmenté significativement les taux d’ET-1 dans les exsudats péritonéaux. Le recrutement des neutrophiles induit par l’ET-1 et l’ovalbumine a diminué chez les souris dépourvues de TNFR1 ainsi que par les traitements ciblant CXCL1/CXCR. Le traitement au bosentan, au BQ-123 ou au BQ-788 a inhibé la production de TNF-‘alfa’ et de CXCL1 induite par l’ET-1 et l’antigène. De plus, la provocation à l’ET-1 et à l’ovalbumine a induit une augmentation des cellules exprimant les marqueurs Gr1+ dans la fenêtre des granulocytes, CD11c+ dans la fenêtre des monocytes, et CD4+ et CD45+ (B220) dans la fenêtre des lymphocytes d’une manière dépendante de l’ETA et de l’ETB, comme déterminé par l’analyse en cytom
