  • Abstract: Exposure to solar radiation can cause unwanted effects, such as photoaging and skin cancer. Thus, different photoprotective approaches are important. The search for new compounds of natural origin with photoprotective potential lead to prospect extreme environments with high radiation levels. This is the case of Antarctic continent, inhabited by organisms capable of protect themselves from UV induced damages. In this study we selected two species of fungi from the Antarctic continent: Geomyces vinaceus, collected from fumarole sediments, and Rhinocladiella similis, isolated from brown algae Phaeurus antarticus, aiming to evaluate the photoprotective potential and antioxidant activity of their extracts. Different cultivation conditions were tested altering culture medium and incubation time to obtain most promising extracts. The extracts were fractionated and subfractionated by vacuum liquid chromatography and thin layer chromatography. Photoprotection was assessed by obtaining the UV absorption spectra and determination of photostability with exposure to UVA radiation. Antioxidant activity was assessed by DPPH radical scavenging and detection of intracellular ROS induced by UVA radiation in cell monolayer (HaCaT). Photoxicity was evaluated by OECD 432 guidelines in mouse fibroblast cell monolayer assay, and ocular irritation was assessed by the HET-CAM assay. The crude extracts of ethyl acetate of fungi cultivated in rice, its fractions and subfractions presented high UVA and UVB absorption, however were considered photounstable under the radiation dose employed. The crudeextract, fractions and subfractions of R. similis cultivated for 14 and 28 days also presented cyto and phototoxic potential, although fractions tested by the HET-CAM assay did not present any irritation potential. Fractions of R. similis cultivated for 28 days presented good scavenging activity of DPPH radical and fractions of R. similis cultivated for 14 days decreased of ROS production induced by UVA up to 22% in lower concentrations (<25 µg/mL). This study contributed to indicate possible applications of fractions and substances isolated from the studied endophytic fungi. However, further studies are needed to confirm the absence of toxicity, evaluate and identify which substances are responsible for cytotoxicity and phototoxicity and which are of interest for photoprotection
