- Abstract: Introduction. The color of oil droplets (ODs) is useful for the determination of the spectral type of photoreceptor since it has been associatede with outer segment absorption properties in some species. Fluorescence of transparent ODs under ultraviolet (UV) ilumination has also been considered as indicative of cone type: in turtle, fluorescent-transparent ODs are thought to be located in blue cones and nonfluorescent ones in UV cones (Vision Res. 34:2801-11, 1994). Objective. To determine prhororeceptor OD types in the hummingbird Eupetomena macroura macroura using fluorescente properties of the ODs. Methods. The eyes were enucleated after 1 hr. in the dark and the retinae dissected from the pigment epithelium. They were placed on slides with physiological solution and immediately examined in a DMRBE Leica epifluorescence microscipe with filter cube A. Results. The retina of this species has a dorsal white field and a ventral yellow field, with clear (or pale) and yellow ODs, respectively. Clear and yellow ODs are the largest in the retina while transparent ones are the smallest. Transparent ODs are autofluorescent in the ventral region and nonfluorescent in the dorsal retina. In addition, both fields have rede an orange ODs. thus the ventral retina contains rede, orange, yellow and fluorescent-transparent ODs . This suggests a total of six photoreceptor types in this retina, four in each region, corresponding to three single cones and one double cone. Conclusion. The color vision of this species presents very clear regional specialization. It may be either tri- or tetrachromatic depending on the size of the stimulus. Since the putative UV cones are in the dorsal retina their role seems to be connected with flower recognition rather than navigation. Accordingly, behavior in a setup which simulates the natural flower feeding situation has shown good discrimination of UV stimuli (IOVS351751, 1994). CNPq, FINEP, FAPESP.
