- Resumo: Ultraviolet light (UV) induces helix-distorting lesions in DNA (Friedberg, 1995). Several studies indicate that one of this lesions, cyclobutane pyrimidine dimers (CPDs), are involved in signaling for UV-induced apoptosis in mammalian cells (Chiganças et al, 2002). However, the role of the damaged DNA replication in this process is not quite clear. To investigate the importance of injured DNA replication in UV-induced apoptosis we utilized aphidicolin, an inhibitor of DNA replication (Spadari, 1984) in wild type and XPB mutated Chinese hamster ovary (CHO) cells expressing a marsupial photolyase (phr gene from Potorous tridactylus) that removes specifically CPDs from the damaged DNA. Our results not only confirmed the efficiency of aphidicolin in blocking DNA synthesis, but also indicated that UV light (UV-C, 254 nm) alone inhibits DNA replication in the hours immediatly subsequent to cells irradiation.In experiments with flow citometry (Fluorescense Activated Cell Sorter-FACS), we were able to quantify the apoptosis ratios in irradiated cells. In these experiments, we demonstrated that cells that had their DNA synthesis blocked after UV are prevented of entering into apoptosis. This result was observed in both wild type and XPB mutated cells, which indicates that the protection observed is independent of NER (nucleotide excision repair). This fact indicates that apoptosis upon UV irradiation requires DNA replicationindependently of the cell proficiency to repair the damaged double-helix
