Immunogenicity of flagelins FliCi e FliCd following oral immunization of mice with attenuated salmonella strains (2003)

- Resumo: The genetic fusion of heterologous epitopes to flagellin expressed by attenuated Salmonella strains represents a promising alternative for the development of bivalent orally delivered recombinant vaccines. In the present work we report the differential immunogenicity of two serological distinct flagellins encoded by the fliCi and fliCd genes following oral immunization of BALB/c immunized with S. Typhimurium (SL3261) or S. Dublin (SL5928) strains. Salmonella strains able to express only one flagellar type were generated following inactivation of the native chromosomal genes and transformation of the mutant strains with plasmids encoding FliCi or FliCd flagellins. The flagellins expressed by each Salmonella strains were in vitro characterized by motility tests and Western blots. Analysis of the FliCi and FliCd immunogenicity were determined in ELISA carried out with sera and fecal extracts of mice orally immunized with the recombinant strains. The results demonstrated that the Salmonella strains were able to express the recombinant, homologous or heterologous, flagellins during in vitro growth but at variable level when compared to the respective parental strains. Moreover, immunization experiments revealed that only the S. Typhimurium strain able to express the FliCi flagellin was able to elicit systemic and secreted anti-flagellin antibody responses following oral delivered of live bacterial cells. The present results suggest that phase I Salmonella flagellinspossess distinct immunological properties in mice following oral administration of attenuated strains. Such observations contribute to the understanding of the remarkable immunological properties of bacterial flagellins and may help to design new bivalent vaccines based on the expression of recombinant flagellins by attenuated Salmonella strains
