- Abstract: The hsp60s are molecular chaperones that guide several steps during protein synthesis, transport and degradation. They are abundant in prokaryotic and eukaryotic cells and highly conserved during evolution. Both the microbial and mammalian hsp60 are the major targets for the immune response against infectious agents. The aminoacid sequence alignment of M. leprae Hsp65 [chaperonin 2] and E. coli HslVU protease suggests two putative threonine catalytic groups, one in the N-domain [Thr136Lys168Tyr264] and the other in the C-domain [Thr375Lys409Ser502]; mutagenesis studies showed that the amino acid residues T375K409S502 at the C-domain form the catalytic group carries out the main proteolytic activity of the M. leprae hsp65. In the present study the putative pathophysiological role of the proteolytic activity of this hsp65 in autoimmune diseases was under evaluation Métodos e Resultados: Experimental autoimmune uveitis [EAU] induced by immunization of the highly susceptible B10.RIII mouse with the Interphotoreceptor Retinoid-Binding Protein [IRBP], and the spontaneous Systemic Lupus Erythematosus [SLE] developed by the [NZB x NZW] F1 interJ strains hybrids were studied. The individuals antiJ IRBP or antiJ nucleus antibody titers were determined, as well as disease scores. Conclusões: The preliminary results suggest that B10.RIII mice intraperitoneally treated with 7 or 14_g of hsp65 develop EAU of a significantly higher incidence and severity thatdoes not correlates with the IgG1 and IgG2a anti? IRBP levels. In SLE, the anti-DNA titers was significantly elevated between of the animal controls and treated with 7_g of hsp65 (P<0,001)
