Melatonin action upon insulin signaling in rat hypothalamus (2003)

- Abstract: Objetivo: Melatonin is the pineal hormone that acts via a pertussis toxin-sensitive G-protein to inhibit adenylate cyclase, however the intracellular melatonin signaling effects are not completely understood. Melatonin receptors are mainly present in the suprachiasmatic nucleus (SCN) and pars tuberalis in both humans and rats. The SCN directly controls the circadian rhythm in plasma glucose concentrations that are increased during the night-time period in pinealectomized rats. In this study, we investigated whether melatonin is able to induce tyrosine and serine/threonine phosphorylation, as well as activation, of high molecular weight insulin signaling proteins in the rat hypothalamus. Métodos e Resultados: The phosphorylation status of the transition preoptic-hypothalamic zone, following melatonin intracerebroventricular infusion, was assessed after immunoprecipitation and immunoblotting with antiphosphotyrosine antibody followed by subsequent immunoblottings with anti-IRS-1, anti-JAK-2, and anti-IR antibodies.) IR kinase activity was measured with ATP incubation after IR immunoprecipitation. The effect was already maximal at a melatonin concentration of 10-8M and occurred 5 minutes after infusion. These results indicate that melatonin has the ability to induce tyrosine phosphorylation in IRS-1, IR and JAK-2 in this hypothalamic area. Conclusões: Since these proteins are involved in the insulin and leptin signaling pathways in this CNS fraction, our resultsdemonstrate a potential cross-talk among these three hormones, and also represent an additional signaling mechanism stimulated by melatonin in the rat hypothalamus in vivo
