- Abstract: AIM: Activation-induced cell death (AICD) is one of the control mechanisms in T cell activation by persistent antigens and an important mechanism of peripheral tolerance. IL-2 is essential for AICD as it enhances expression of Fas ligand (FasL) and decreases the expression of its inhibitor FLIP. Common variable immunodeficiency (CVID) is a heterogeneous group of immunologic disorders of unknown etiology, characterized by impaired antibody responses. This study investigates the role of AICD in the pathogenesis of CVID. Material and Methods: Blood samples were collected from CVID patients and the mononuclear cells were separated by gradient centrifugation. Peripheral blood mononuclear cells (PBMC) were subjected to flow cytometry (FACS) to analyze expression of CD3, CD3/CD4 and CD95 markers. PBMC were cultured with phytohemaglutin A (PHA) during 72 hours and AICD was analyzed by Anexin-V-FITC staining.Results: We found that a subgroup of CVID patients with lymphopenia had up regulated CD95 expression and an increased rate of apoptosis when PBMC were stimulated in vitro with PHA. Discussion: Our data suggest that lymphopenia in these patients may be associated with increased apoptosis probably due to up regulation of Fas-mediated activation-induced cell death in T cells
