- Abstract: Aims: MAM makes a bridge between lymphocytes T receptor and I-Ealpha MHC class II molecules. Here we studied if MAM-induced nitric oxide (NO) production is H-2 restricted, dependent on the lymphocyte, IFNg or TLR4 signaling. Methods: Resident peritoneal cells (4x105 cells/100 mL) from C3H/HePas (H-2k), C3H/HeJ (H-2k), BALB/c (H-2d), C57BL/6 (H-2b) mice and J774A.1 cells (H-2d - 1x105 cells/100 mL) were co-cultured in the presence or absence of spleen lymphocytes (1:2 ratio) and stimulated with MAM or LPS (1 microg/mL). Nitrite concentration was measured in culture supernatants by Griess reaction. Results: In peritoneal cells from C3H/HePas, C3H/HeJ and BALB/c, maximal NO release was obtained with 1/100 dilution of MAM and 48 h incubation being around 75 microM. Significantly lower levels were found in C57Bl/6 peritoneal cells (18.18 microM). LPS potentiated MAM-induced NO production in C3H/HePas, but not in C3H/HeJ or C57Bl/6 mice (strain non-responsive to LPS or MAM, respectively).Treatment of peritoneal cell with antibody to IFNg inhibited dose-dependently NO production. Depletion of non-adherent cells significantly decreased NO production (60 microM vs 1.04 microM) and addition of IFNgamma potentiated MAM induced NO production. In J774A.1 macrophages MAM induced NO release but only in the presence of viable BALB/c spleen lymphocytes were added (4.24 vs 28.85 microM). Conclusion: NO production induced by MAM is a) restricted toH-2K and H-2d mice strains; b) independent of TLR4 signaling; c) potentiated by LPS; d) dependent on the presence of non-adherent peritoneal cells or viable spleen lymphocytes; e) regulated by IFNg
