- Abstract: Objetivo: Physical exercise influences brain trophism and plasticity. Moderate treadmill running triggers changes on the apomorphine-induced circling behaviour in the 6-hydroxydopamine (6-OHDA) model of Parkinsonism as well as on motor parameters detected by infrared motion sensor. In the present work we have investigated the effects of moderate treadmill running on degenerative events in the nigrostriatal system after striatal injection of 6-OHDA. Métodos e Resultados: Rats were submitted to 5 days per week session in the treadmill: 6m/min for 20 min (first week) and 12m/min for 30 min (second week); rats trained 12m/min for 45 min in the third week, a regime that was maintained after surgery. After 6 weeks, trained and sedentary rats were submitted to unilateral striatal injection of 6-OHDA or solvent. The rats were sacrificed at 21 days and 50 days after dopamine lesion.) After sacrifice, rat brains were processed for tyrosine hydroxylase, glial fibrillary acidic protein, OX-42 immunohistochemistry, markers for dopamine cells, astrocytes and microglia, respectively, and quantified using stereology. The volume of the striatal lesion and the microglia activation was not changed in the trained rats compared to the sedentary rats in the two time-points. However, the volume of the astroglial reaction was decreased by 39% (p<0.01, t test) in the striatum of the trained rats. Conclusões: The results indicate that moderate treadmill running does not protectstriatum from 6-OHDA induced dopamine nerve terminal disappearance but may regulate later changes in astroglial reaction
