Sequence analysis and molecular cloning of the Streptococcus mutans oligopeptide permease (2003)

- Abstract: The recent advent of the genome sequencing technology and the availability of bioinformatic tools had a tremendous impact on the identification of genes involved in pathogenicity and different metabolic features among different bacterial species. In this work we investigated the presence of the major oligopeptide uptake (Opp) system among eubacteria and archea with genomes available at the Genebank. Moreover, we examined the genetic organization and performed similarity analysis of the opp genes found in the genome of the gram-positive pathogen Streptococcus mutans, one of the main etiologic agents of tooth decay in humans. The opp operon was found in 24,5% (29/119) of the eubacteria genomes reported so far, while among archea species the opp operon was found in 5,5% (1/19) of the genomes available at the Genebank. The S. mutans opp genes are arranged in an operon-like organization including five cistrons: oppA, encoding the peptide binding domain, oppB and oppC, encoding membrane proteins responsible for the uptake of substrate peptides, and oppD and oppF, encoding the ATP-binding components which provide energy for the active transport of peptides to the bacterial cell. Comparison of the amino acid sequences of the S. mutans opp genes with other gram-negative and gram-positive bacterial pathogens were carried out and allowed the definition of phylogenetic trees of each Opp system component and identification of functional and structural domains. Finally the S. mutansoppA gene was amplified and cloned in E. coli K12 plasmid expression vectors. Identification and cloning of Opp proteins will contribute to the understanding of the physiological role of the Opp system on the nutrition strategies and pathogenicity of S. mutans
