The hyperthyroidism and the renin-angiotensin system on cardiac microcirculation (2004)

- Abstract: Objetivo: The aim of this study was to determine the role of renin-angiotensin system (RAS) in the capillaries growth in a model of cardiac hypertrophy induced by thyroxine (T4). Métodos e Resultados: Wistar adults rats (n=5 per group) were divided as follow: T4-100 (0.1 mg/kg/day, i.p.), T4-25 (0.025 mg/kg/day, i.p.), ENA (Enalapril, 250 mg/L/day, orally), LOS (Losartan, 200 mg/L/day, orally), T4-100+ENA and T4-100+LOS. Body weight and hemodynamics parameters were obtained. After 7 days of treatment, the rats were anesthetized, hearts were perfused with saline, KCl (14 mM) and paraformaldehyde. Left ventricles were processed to histological analysis. Capillary densities (CD; number of capillaries per square millimeter) and capillary/fiber ratio (CFR) were determined. Results: There was a significant decrease in the CD on the T4-100 group compared to control group (733.32 ± 72.13 vs. 1079.8 ± 41.13; P<0.05) despite a significant increase of 33% on myocardial mass. Then, the CFR was significantly lower in T4-100 group (0.59 ± 0.085 vs. 0.79 ± 0.06 to control group, P<0.05). The use of Enalapril on hyperthyroid animals was efficient to prevent the cardiac hypertrophy, but did not affect the angiogenic response compared to T4-100 group. On the other hand, the administration of Losartan on hyperthyroid rats did not alter the myocardial mass but promoted a significant increase on CD (1070.6 ± 158 vs. 733.3 ± 72.1 to T4-100 group). Conclusões: Considering thatthe T4 promoted a more intensive increase on myocardial mass than the decrease promoted on CD, our data suggest an important angiogenic effect of the hormone. In addition, the use of both RAS inhibitors in control or hyperthyroid rats promoted different results on cardiac microcirculation
