Effect of allergen sensitization on eosinophil precursor responses to cytokines and chemokines in bone-marrow culture (2005)

- Abstract: We previously described modulation of bone-marrow (BM) responses to eosinopoietic cytokines by allergen sensitization and chalenge, as well by dexamethasone (DEX), prostaglandin E2 (PGE2), and indomethacin (INDO). Here we evaluated whether these factors interact in IL-5 stimulated BM cultures, and whether this affects responses to IL-13 and Eotaxin. Methods and Resulted: We evaluated the modulation of eosinophil precursor responses to IL-5 in liquid BM culture by different agents, comparing naive and allergic BALB/c and C57BL/10 mice. C57BL/10 responded to allergen sensitization and challenge differently from BALB/c mice. Comparable responses to DEX and PGE2 were observed in both strains. Indomethacin was effective in naive but not immune BALB/c mice, and ineffective in C57BL/10 mice. We analysed the response to IL-13, a cytokine that plays a central role in the pathophysiology of asthma. IL-13 synergized with IL-5 in stimulation of bone-marrow from naive BALB/c mice, but inhibited dose-dependently the response to IL-5 in sensitized and challenged mice. On the other hand, IL-13 was ineffective on BM from naïve C57BL/10 mice. By contrast, it significantly inhibited responses to IL-5 in bone-marrow of sensitized/challenged mice of the same strain. In the BALB/c strain, sensitization followed by airway challenge shifted eotaxin from a strongly synergic, stimulatory agent, to a powerful inhibitor of the same response, pointing to immunomodulation ofbone-marrow response patterns. By contrast, C57BL/10 mice were refractory to eotaxin, regardless of sensitization, indicating that the pattern of response to eotaxin depends on the genetic background. Conclusions: Bone-marrow responses to different modulatory agents depend on at least 3 factors: mouse strain, the immunological status of the host and the agent itself
