Influence of renin angiotensin system (RAS) in the thyroid hormone-induced cardiac hypertrophy. Study using primary cultures of neonatal cardiomyocytes (2005)

- Abstract: Objetivo: We have previously demonstrated in vivo the participation of RAS in the thyroid hormone-induced cardiac hypertrophy. However, the contribution of local RAS for this effect has not been evidenced yet. Then, the objective of present study was to investigate the role of local RAS to the development of cardiac hypertrophy induced by T3, using primary cultures of cardiomyocytes. Métodos e Resultados: Cultures were prepared from neonatal ventricles of Sprague Dawley rats by colagenase/pancreatin method. Myocytes were segregated using Percoll density gradient and cultured in DMEM containing serum, which was changed to serum-free medium after 24 hours. Cells were treated with T3 (10nM), T3 plus Losartan (0.3µM) and/or T3 plus Enalapril (1µM) for 24h. Total RNA was isolated by Trizol, and cDNA was obtained. The Atrial Natriuretic Factor (ANF) and Alpha-Smooth Muscle Actin (Alpha-SM Actin) mRNAs were employed like cardiac hypertrophy markers, using specific primers to PCR. The results were expressed as mean ± SD and P<0.05 (n=3). T3 promoted a significant cardiac hypertrophy, expressed in % by increased ANF and alpha-sm actin mRNA levels. The cardiac hypertrophy was prevented partially by administration of losartan or by enalapril. Conclusões: These results indicate that the local (cardiac) RAS has an important role in the development of thyroid hormone-induced cardiac hypertrophy
