Modulation of kinin receptors expression by AT1 receptor activation in hypertensive rats (2005)

- Abstract: Objetivo: Kinins participate in various physiological and pathological processes, including vasodilation, release of endothelium derived factors and inflammation. Their biological actions occur through activation of G-protein coupled receptors, named B1 (inducible) and B2 (constitutive). To investigate the role of angiotensin II on the expression of kinin receptors in two models of experimental hypertension. Métodos e Resultados: Studies were performed in male adult 2 Kidneys-1 Clip (2K-1C) and Spontaneoulsy Hypertensive Rats (SHR), compared to normotensive Wistar rats (n=7). Animals were treated with losartan (AT1 receptor antagonist; 15mg/kg/ 7 days). Blood pressure (BP) was measured by tail pletismography. Cardiac mRNA levels for B1 and B2 receptors wee determined by Real Time PCR. B1 receptor localization was performed by immunohistochemistry. Hearts of 2K-1C rats (BP 195±7 mmHg) presented a 15-fold increase in B1 and a 4-fold increase in B2 mRNA levels in comparison to control (BP 122±3 mmHg). In contrast, no difference was observed on receptors expression of SHR (BP 184±4 mmHg) vs control (BP 118±2 mmHg). In 2K-1C rats, B1 receptor was localized in cardiomyocytes membrane and in the endothelium of cardiac microvessels. Losartan treatment reverted B1 protein expression and B1/B2 mRNA levels (*P<.05), despite reducing, but not normalizing BP (139±5 mmHg*). Conclusões: Our results provide strong evidence that, in a renin-dependent model of hypertension, angiotensin IImodulates the expression of kinin receptors. This phenomenon might not be related to high blood pressure levels. These data reveal a novel site of interaction between kinins and angiotensin II
