- Abstract: Negative feedback-loops, i.e. the inhibition of the expression of circadian clock genes by the corresponding proteins after a certain time lag of delay, are at the core of all current models of circadian clocks. In particular at the physiological and metabolic level it has been shown that multi-loop negative feedback systems exhibit more robust dynamics with respect to noise and parameter variations. Our intention in this work is to check whether this also applies to molecular circadian systems, in particular under external periodic stimuli. We therefore extend a minimal, yet realistic model for gene expression oscillations, consisting of one delay differential equation, by adding a second feedback loop.We numerically investigate the sensitivity to parameter changes, and the responses to periodic external stimuli for diverse combinations of inhibition thresholds and delay times. Our findings confirm the increase of robustness by addition of a second feedback loop. For instance, the range of external driver periods leading to stable entrainment is expanded, and chaotic dynamics being observed in the single-loop system under contradictory stimulation with two external phase-shifted drivers are converted to frequency-locked, periodic dynamics through the addition of a second loop. With these examples, we will discuss the putative role of multiple, redundant, feedback loops for the function and evolution of circadian pacemakers.
