- Abstract: Introduction and Objectives: Complement and dendritic cells (DCs) are essential components of innate immunity. Both participate in local inflammation and moreover have roles in the initiation of the acquired immunity response and in the maintenance of tolerance. Recent studies have demonstrated the ability of DCs to synthesize C1q, C3, Factor I, Factor B and complement receptors CR3 and CR4. In this study we investigated if DCs are a source of others complement proteins. Methods and Results: DCs were generated in vitro from leukocytes obtained by leukapheresis from healthy donors. Adherent peripheral blood mononuclear cells were isolated and cultured in the presence of GM-CSF plus IL-4 in culture medium supplemented with fetal calf serum. DCs obtained by day 7 were CD14-CD1ahigh, CD11c+HLADR+ and had typical DC morphology. For the activation of DCs, LPS was added in the last 24 h of culture, causing an increase in the expression of co-stimulatory molecules. We demonstrate that human DCs are a source of other soluble complement proteins including C1q, C4b binding protein (C4BP), C7 and C8. Complement receptors (CR)1 and the CD18 chain (common for CR3 and CR4) were also present on DCs while CR2 was not detected. Conclusion: Human monocyte-derived DCs produce complement components, regulators and receptors.
